Immune Dysregulation and Cytokine Circuitry in Genital Endometriosis: Mechanistic Insights and Next-Generation Immunotherapeutic Strategies
Abstract: Genital endometriosis is increasingly recognized as an immune-mediated inflammatory disorder driven by complex interactions between dysregulated immune cells, cytokine hubs, and microbiome-derived modulators. This review introduces a novel “immune–cytokine circuitry” framework that unifies innate and adaptive immune abnormalities with key cytokine loops sustaining chronic inflammation, angiogenesis, neuroinflammation, and immune tolerance. Within this circuitry, macrophage polarization, dendritic cell immaturity, NK-cell anergy, Treg expansion, Th17 amplification, and B-cell–mediated autoimmunity converge to establish …

Vitamin D Mitigates Inflammation and Downregulates Importin α3 in Non-Alcoholic Fatty Liver Disease (NAFLD)
Abstract: Background: Pro-inflammatory cytokines, such as TNF-α, IL-1β, IL-6, and IL-8, seem to play a crucial role in the progression of NAFLD as they activate the transcription factor NF-кB. The activated NF-кBp50/RelA subunits are translocated to the nucleus by Importin α3 and Importin α4. Numerous studies have indicated a negative association between NAFLD and vitamin D levels. Low vitamin D levels have been correlated with histological severity, necro-inflammation, and fibrosis in …

Drug Induced Immune Mediated Nephritis: Molecular Mechanism , Pathways and Clinical Implications
Abstract: Drug-induced immune-mediated nephritis (DI-IMN) has become a more widely known cause of acute kidney injury (AKI), with the potential for development to chronic kidney disease if not detected and treated promptly. T-cell hypersensitivity to pharmaceuticals, such as antibiotics, proton pump inhibitors, nonsteroidal anti-inflammatory drugs, and immunological drugs, are the major causes for it. Vitamin D Mitigates Inflammation and Downregulates Importin α3 in Non-Alcoholic Fatty Liver Disease (NAFLD)
Abstract: Non-alcoholic fatty liver disease (NAFLD) affects about one-third of the population in the United States and Western countries, reaching up to 90% among obese individuals and those undergoing bariatric surgery. Characterized by hepatic fat accumulation, NAFLD progresses from simple steatosis to non-alcoholic steatohepatitis (NASH), fibrosis, and, in rare cases, hepatocellular carcinoma. Linked intricately with metabolic syndrome, including diabetes and obesity, NAFLD development involves insulin resistance and hyperinsulinemia. Experimental evidence consistently …

Rheumatoid Arthritis and Stem Cell Therapies Perspectives for It
Abstract: Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by systemic inflammation, primarily affecting synovial joints, leading to joint destruction and systemic complications. The pathogenesis involves immune system dysregulation, autoantibody production, and enzymatic imbalances. Enzymes, including matrix metalloproteinases (MMPs), serine proteases, and kinases, play crucial roles in these processes. Their dysregulation contributes significantly to disease progression through the imbalance of pro-inflammatory cytokines, oxidative stress, and extracellular matrix (ECM) degradation. MMPs, …
