Application of Prodrug to Improve Skin Permeability: A Review
Abstract: Administration of drug via skin has some limitations such as skin barrier, solubility of drug and partition coefficient etc. Stratum corneum which is an outer skin layer, acts as a barrier to the skin. Thus, prodrug has some applications to improve skin permeability. Prodrugs are defined as the compounds which are initially inactive but covert into their active form when reacted with several chemicals or enzymes at particular active site …

Review: Prodrug Concept in Drug Design
Abstract: AbstractIn the world of drug discovery and development, prodrugs have become a powerful method for improving biopharmaceutical, physicochemical or pharmacokinetic properties of pharmacologically active agents. The purpose of this review is to provide some types of prodrugs which are bioreversible derivatives of drug molecules that undergo an enzymatic and/or chemical transformation in vivo to release the active parent drug, which can then exert the desired pharmacological effect. The target of …

Biopolymer-Based 3D Composite Scaffolds for Bone Tissue Engineering: Photothermal-Responsive Systems with Controlled Pt (IV) Prodrug Release
Abstract: Biopolymer-based composite scaffolds have attracted significant attention as multifunctional platforms at the intersection of polymer science, regenerative medicine, and cancer nanotherapy. In this study, a dual-functional scaffold was developed for simultaneous osteosarcoma treatment and bone regeneration using poly (L-lactic acid) (PLLA) reinforced with bioactive glass (BG) and integrated with a glutathione-responsive platinum (IV) prodrug system (PDA@Pt). To improve interfacial adhesion, photothermal conversion efficiency, and tumor-specific activation, the Pt (IV) prodrug …

Prodrugs and Its Types
Abstract: In this study, description of prodrugs is given. Prodrugs are biologically inactive agents that upon biotransformation in vivo result in active drug molecules. Inactive prodrugs are pharmacologically inactive medications that are metabolized into an active form within the body. Depending upon the functional group, used to design the produrg, present in the parent drug, the pro drugs can be classified so many ways, which are given in this study. Keywords: …

Drugs and Polymers Used for Colon Specific Drug Delivery System with Current Trends: A Systematic Review
Abstract: Due to greater flexibility in design of dosage form and high patient compatibility, oral route of administration is the most preferred route but the gastrointestinal tract has many types of other undefeatable barriers to drug delivery. In oral colon-specific drug delivery system, colon has a large amount of lymphoma tissue which facilitates direct absorption into the blood, negligible brush border membrane activity, and much less pancreatic enzymatic activity as compared …

Aceclofenac and Diclofenac Sodium Conjugates with Amino Acids to Overcome Acidity
Abstract: This research work was based on a conjugate approach with some non-steroidal anti-inflammatory drugs (NSAIDs) to overcome the side effects like gastric irritation, solubility behavior, and some other effects of NSAIDs. Almost all the NSAIDs have undesirable reactions as well as they cause some types of major adverse reactions. Here the prodrugs conjugates are the approaches that increase the lipophilicity of the drug and decrease the solubility in water by …

Synthesis, Characterization and Pharmacological Evaluation of Amide Derivates of Prednisolone
Abstract: prodrugs. These novel prodrugs were also characterized with the help of usual analytical and spectral techniques (IR spectroscopy and NMR spectroscopy). These newly synthesized ester derivates of prodrugs were screened for their spectral techniques. All the above compounds showed significant anti-inflammatory effect at 50 mg/kg p.o. and the experimental data are statistically significant at p< 0.05 level. The pharmacological activities exhibited by synthesized novel compounds have confirmed that they may …

Comprehensive Overview of Carbohydrate Polymers
Abstract: Targeting drug delivery specifically to the colon has become increasingly crucial for treating various colonic diseases like colorectal cancer, amebiasis, ulcerative colitis, and Crohn’s disease. Numerous strategies have been developed for this purpose, including using enzymatically degradable polymers, employing prodrug-based approaches, coating drugs with polymers sensitive to time or pH changes, utilizing osmotically and pressure-controlled delivery systems. Polysaccharides activated by the colon's physiological environment show significant potential as they offer …

Recent Trends in Ocular Drug Delivery: Challenges and Approaches
Abstract: Drug topically regulation can be accessed most easily in the eye. When given topically as eye drops, medication ocular bioavailability is quite low. Drug entry into specific ocular locations is complicated by the complex anatomy and physiology of the human eye. Research has long been interested in the topical administration of successful treatments. To provide a suitable ocular penetration and extend the duration of drug residence is their challenging task. …

Modified Nucleosides “(ProTide)†as Potential Anti-HCV Therapeutics
Abstract: Hepatitis C virus (HCV) is among the most common causes of cirrhosis and chronic liver disease worldwide. As a result, many researchers are interested in designing and synthesis of a clinical treatment for HCV. Nucleoside monophosphates and monophosphonates play such an important role for treatments of incurable diseases such as hepatitis. For example, 2'-C-methyladenosine and 2'-C-methyl guanosine have shown activities against HCV in the replicon assay as well as against …
