Design and Evaluation of Promethazine Hydrochloride Rapimelts

Abstract

Objective: In the present study, efforts were taken to develop rapimelts of promethazine HCl—an antiemetic agent with an objective to achieve rapid disintegration, and further improve thebioavailability of drug.Methods:Rapimelts were prepared by direct compression methodusing superdisintegrants such as crospovidone (cp), croscarmellose sodium (ccs), sodiumstarch glycolate (ssg). The prepared formulations were evaluated for precompressionparameters such as bulk density, tapped density, Carr’s compressibility, Hausner ratio andangle of repose. The prepared batches of rapimelts of promethazine HCl were evaluated forhardness, weight variation, thickness, friability, drug content, disintegration time, wettingtime, water absorption ratio, andin vitrodissolution profile.Results: From the FourierTransform Infrared Spectroscopy (FTIR) studies, the compatibility of drug-polymers wasconfirmed. All the rapimelts were shown satisfactory results which complies with officiallimits. Among the prepared nine formulations, the formulation containing 7.5% crospovidone(PF9) showed highest drug release of 98.6% than other formulations.Conclusion:From thisstudy we can conclude that formulated rapimelts of promethazine HCl containingcrospovidone are better and effective than conventional tablets to meet patient complianceand give fast relief from vomiting and emesis.Keywords: Promethazine HCl, rapimelts, superdisintegrantCite this ArticleSelvam RP, Gowrav MP, Chandy V.Design and Evaluation of PromethazineHydrochloride Rapimelts.
