The Dual Crisis: Antibiotic Resistance and the Discovery Void – A Review of Novel Therapeutic Strategies and Non-Traditional Approaches
Abstract: The global rise of antimicrobial resistance (AMR) has emerged as one of the most critical public health challenges of the 21st century, threatening to undermine decades of therapeutic success and rendering conventional antibiotic regimens increasingly ineffective. Parallel to the escalating resistance rates is a profound “discovery void,” characterised by a steep decline in the development of new antibiotic classes since the late 20th century. Together, these two interconnected crises create …

Emerging Therapies in Respiratory Tract Infections: Current Advances and Future Perspectives
Abstract: The impact of respiratory tract infections (RTI) as a global public health problem continues to be high morbidity and mortality rates. The development of novel therapeutic approaches stemmed from the fact that conventional treatment strategies, antibiotics, antivirals, antifungals etc. have become increasingly ineffective because of rise of antimicrobial resistance (AMR). Next generation therapies, such as novel antimicrobial agents, host directed therapies and novel drug delivery systems have come on the …

Quorum Sensing and Biofilm Dynamics in Pathogens: An Evolving Frontier in Infectious Disease
Abstract: Bacterial and fungal pathogens rarely act as solitary invaders. Instead, they engage in sophisticated chemical communication known as quorum sensing (QS) to coordinate population-wide behaviours and construct structured, multicellular biofilm communities. These biofilms, composed of extracellular polymeric substances, not only facilitate microbial persistence on biotic and abiotic surfaces but also confer remarkable protection against host immune defenses and antimicrobial therapies. QS systems act as central regulators of biofilm dynamics, orchestrating …

Investigating the Impact of Viral Infections on Pharmacokinetics and Pharmacodynamics of Antiviral Drugs
Abstract: The emergence and re-emergence of viral infections pose significant challenges to public health, necessitating a comprehensive understanding of how these infections impact the pharmacokinetics and pharmacodynamics of antiviral drugs. This review explores the intricate interactions between viral pathophysiology and the pharmacological effects of antiviral agents. Factors such as viral replication dynamics, host immune responses, and the genetic variability of both viruses and hosts significantly influence drug absorption, distribution, metabolism, and …

Toxicological Profiling and Safety Assessment of NASAT 2.0: A Conceptual Framework for Adaptive Nanoparticle Therapy in Rabies
Abstract: Rabies, caused by the highly neurotropic Rabies lyssavirus, remains one of the most enigmatic and universally lethal infectious diseases known to modern medicine. Once clinical symptoms manifest following successful neuroinvasion, the fatality rate approaches absolute certainty (approximately 99.9%), a staggering statistic that has remained largely unchallenged despite massive, concurrent advances in modern virology, critical care medicine, and cellular immunology. Current late-stage therapeutic protocols, most notably the widely debated Milwaukee Protocol, …

Plant Bioactive Metabolites and Their Role in the Prevention and Control of Dental Diseases and Infections.
Abstract: The oral diseases, including dental caries and periodontitis, affect masses worldwide, and have historically been managed with synthetic antimicrobials that cause deleterious side effects, leading to mounting bacterial resistance. Consequently, contemporary dentistry is rapidly shifting toward phytomedicine using plant bioactive molecules, such as polyphenols, terpenes, anthraquinones (berberin, eugenol, curcumin, catechins, allicin, and azadirachtin. These compounds offer potent antimicrobial, anti-inflammatory, and tissue-regenerative properties that are crucial for preventing and curing dental …
