Abstract

Urolithiasis is a complex process of kidney stone formation involves various stages of nucleation and mineralization. Plants are used for treatment of urolithiasis from early times. Thus the study was evolved around the plant Rotula aquatica Lour. used as “Paashanabhedha”. The present study reveals the bioactivity guided isolation of anti-urolithiatic compounds from successive solvent extracts of the roots of Rotula aquatica Lour. The isolated compounds showed significant in-vitro antiurolithiatic activity compared to Cystone®. The active aqueous extract was partitioned by column chromatography. The active 4th fraction was re-chromatographed to 12 sub-fractions. The active 7th and 8th sub-fractions were combined and subjected to prep-TLC. Two compounds, RA-01 and RA-02 had significant activity % inhibition of 45.21, 46.58, 49.74% and 46.53, 49.57, 54.37% at 0, 60, 120 minutes respectively. The results suggest the dose depended inhibition of crystal formation. Compounds were characterized and structure was elucidated as class of phenolic compounds: gallic acid and p-coumaric acid by spectral studies. The probable mechanisms include hydrolysis of calcium oxalate and calcium phosphate in presence of –OH of the isolated compounds. Thus  the study proves traditional use of Rotula aquatica Lour. in treatment of kidney stones. Keywords: Rotula aquatica Lour., anti-urolithiatic, in-vitro bioactivity guided isolation, characterization, gallic acid, p-coumaric acidCite this ArticlePrashanthi P.
