Preparation and Evaluation of Mucoadhesive Bilayered Buccal Patch using HPMC and Gelatin
Abstract
The buccal drug delivery system has gained more popularity than other drug delivery systems owing to its ease and safe administration. The high permeability of the buccal mucosa allows the drug to reach the systemic circulation and gives rapid onset of action. The buccal route also helps to avoid drug degradation from first-pass hepatic metabolism and improves drug bioavailability. The present study is designed to develop and evaluate the mucoadhesive buccal patches using HPMC (Hydroxypropyl Methyl Cellulose) and gelatine. The patches were prepared by the solvent casting method. Propylene glycol (PG) and Dimethyl sulfoxide (DMSO) was used as plasticizer and permeation enhancer, respectively. HPMC was used as a mucoadhesive polymer. The developed bilayered patch was evaluated for thickness, weight, moisture content, folding endurance, moisture uptake, etc. The drug content uniformity, drug permeability, and drug release study from the patches was investigated using ascorbic acid as a model drug. The results indicated that the bilayered buccal patch can be prepared successfully using HPMC as a mucoadhesive polymer with adequate mucoadhesion. Keywords
