Estimation of Effective Storage Time in Post-thawing and Washing Cryopreserved Red Blood Cells using Mechanical Properties of Membrane by Dielectrophoresis

Abstract

Aim: The aim of the study is to estimate most effective storage time after thawing and washing in cryopreserved red blood cells(RBCs), using the mechanical properties of red blood cells(RBCs) membrane by dielectrophoresis. Materials and Methods: The mechanical properties of the RBCs membrane(elasticity, electric capacity and interactive energy) were calculated by mathematical model made from dielectrophoresis on RBCs. In addition, estimating the most effective storage time after thawing and washing in cryopreserved red blood cells(RBCs), the conventional quality variables of cryopreserved RBCs including osmotic resistance, globular RBCs, adenosine triphosphate (ATP) and 2,3-diphosphoglycerate acid (2,3-DPG) concentration in red blood cells(RBCs), and supernatant free Hb were evaluated. Results: Statistically significant changes were observed in the mechanical properties of the cryopreserved red blood cell membrane after thawing and washing from 24 hours of storage compared to pre-frozen and 21 days stored ACD(adenine citrate dextrose at 4℃) blood, while osmotic resistance did not differ significantly as compared to pre-frozen and ATP concentration as compared to 21 days stored ACD blood. Statistically significant changes were observed in the The globular RBCs and supernatant free Hb after thawing and washing from 36, 48 hours of storage compared to 21 days stored ACD(adenine citrate dextrose at 4℃) blood respectively, while adenosine triphosphate (ATP) and 2,3-diphosphoglycerate acid (2,3-DPG) concentration in red blood cells(RBCs) did not differ significantly. Conclusion: The mechanical properties of RBCs membrane by dielectrophoresis can be used for observing biophysical characters of cryopreserved RBCs and the most effective storage time of cryopreserved red blood cells after thawing and washing is 24 hours.
